Descrição: Esta discipliina tem como objetivo apresentar os mecanismos envolvidos na diferenciação de células-tronco envolvidas na formação do dente e dos tecidos de suporte, assim como de células envolvidas no catabolismo de tecidos mineralizados.

Conteúdo programático:

1) Cultura de células eucarióticas
- Tipos de culturas: células primárias e linhagens celulares.
- Modo de crescimento: células aderentes ou em suspensão.
- Meios de cultivo e suplementações utilizadas.
- Avaliação microscópica das células em cultura.
- Congelamento, descongelamento e armazenamento celular.
- Contaminação da cultura: uso de antibióticos e antifúngicos.

2) Métodos laboratoriais para detecção de ácidos nucleicos, proteínas e enzimas
- Análise do DNA e da expressão gênica (RNA): reação em cadeia da polimerase (PCR) em tempo real e silenciamento de RNAm.
- Análise da síntese de proteínas intra e extra-celulares: ensaio imunoenzimático (ELISA), western blotting, imunocitoquímica, imunohistoquímica, imunofluorescência.
- Análise da atividade enzimática: fosfatase ácida resistente ao tartarato (TRAP), zimografia convencional, zimografia in situ.

3) Pesquisa translacional
- Modelos animais utilizados em pesquisas pré-clínicas em Odontologia.
- Papel dos mediadores inflamatórios na inflamação e no reparo ósseo e dentinário.
-Reguladores da osteoclastogênese, reabsorção óssea e reabsorção dentária como potenciais estratégias terapêuticas inovadoras.
- Modelos in vivo e ensaios funcionais para investigação do processo de biomineralização.
